Abstract: Thus, we apply a frequency-domain acoustic full-waveform inversion (FWI) method to obtain high-resolutionWe discuss the influence of the frequency group selection strategy and the tunnel observation systemBased on the conventional strategies of frequency-domain acoustic FWI, we propose a frequency group selectionstrategy that combines a low-frequency selection covering the vertical wavenumber and a high-frequencyThis strategy can effectively obtain the spatial structure and physical parameters of the geology ahead

Biological chip technology to quickly batch select optimum cryopreservation procedure

YU Lina, LIU Jing, ZHOU Yixin, HUA Zezhao

Frontiers in Energy 2007, Volume 1, Issue 3,   Pages 316-321 doi: 10.1007/s11708-007-0046-2

Abstract: In the practices of cryobiology, selection of an optimum freeze/thawing program and an idealistic cryoprotective agent often requires rather tedious, time consuming and repetitive tests. Integrating the functions of sample preparation and viability detection, the concept of biochip technology was introduced to the field of cryopreservation, aiming at quickly finding an optimum freezing and thawing program. Prototype devices were fabricated and corresponding experimental tests were performed. It was shown that microflow-channel chip could not offer a high quality solution distribution. As an alternative, the spot-dropping chip proved to be an excellent way to load the sample quickly and reliably. Infrared thermal mapping on such a chip showed that it had a rather uniform heat transfer boundary. Applying the spot-dropping chip combined with the thermoelectric cooling device, the final output of cryopreservation of multiple samples was tested, and the optimal freeze/thawing program as well as the potentially best concentration of the cryoprotective agent was found by analyzing the results. Further, application of this technique to measure the thermo-physical properties of the cryo-protective agent was also investigated. The study demonstrated that a biochip with integrated automatic loading and inspection units opens the possibility of a massive optimization of the complex cryopreservation program in a quicker and more economical way.

Frontiers of Structural and Civil Engineering doi: 10.1007/s11709-023-0006-x

Abstract: A frequency error estimation is presented for the isogeometric free vibration analysis of Kirchhoff–LoveBy analyzing the discrete isogeometric equations with the aid of harmonic wave assumption, the frequencyIn particular, the governing relationship of the continuum frequency for Kirchhoff–Love cylindrical shellsis naturally embedded into the frequency error measures without the need of explicit frequency expressionsNumerical results not only thoroughly verify the theoretical convergence rates of frequency solutions

Potential unreliability of ALK variant allele frequency in the efficacy prediction of targeted therapy

Frontiers of Medicine 2023, Volume 17, Issue 3,   Pages 493-502 doi: 10.1007/s11684-022-0946-x

Abstract: Anaplastic lymphoma kinase (ALK) is the most common fusion gene involved in non-small cell lung cancer (NSCLC), and remarkable response has been achieved with the use of ALK tyrosine kinase inhibitors (ALK-TKIs). However, the clinical efficacy is highly variable. Pre-existing intratumoral heterogeneity (ITH) has been proven to contribute to the poor treatment response and the resistance to targeted therapies. In this work, we investigated whether the variant allele frequencies (VAFs) of ALK fusions can help assess ITH and predict targeted therapy efficacy. Through the application of next-generation sequencing (NGS), 7.2% (326/4548) of patients were detected to be ALK positive. On the basis of the adjusted VAF (adjVAF, VAF normalization for tumor purity) of four different threshold values (adjVAF < 50%, 40%, 30%, or 20%), the association of ALK subclonality with crizotinib efficacy was assessed. Nonetheless, no statistical association was observed between median progression-free survival (PFS) and ALK subclonality assessed by adjVAF, and a poor correlation of adjVAF with PFS was found among the 85 patients who received first-line crizotinib. Results suggest that the ALK VAF determined by hybrid capture-based NGS is probably unreliable for ITH assessment and targeted therapy efficacy prediction in NSCLC.
